Next, we tried it on Feraheme, a FDA-approved iron oxide nanodrug, tracked by [89]Zr PET. One 15-minute priming step with MPN nearly doubled tumor uptake at 6 hours and held onto a ~30% gain out to 24. Same principle, clinical-grade cargo.

So does vasodilation also mean more nanoparticle delivery? Give a 70 kDa dextran 15 minutes after MPN and the tumor takes up far more of it (~ 1.5×) and crucially, it goes also more into the core rather than piling up at the rim where most nanomedicine gets stranded.

And NitricO means vasodilation. Using optoacoustic imaging (RSOM), we showed tumor vessels widening in real time. The striking part: tumors responded more than three times as strongly as healthy skin. The messy, leaky vasculature we usually curse turned out to be the most responsive.

Meet MPN = methyl palmitate nanoparticles, made by Paolo's group. Simply an albumin shell wrapped around a lipid your own body already makes. Paolo's group had already shown they stun macrophage activity over a few hours (PMID 34617542). Our new paper is about something much faster.
