SIRPG KD enhances effector cytokines and granzyme production in CD45RO+ CD8 T cells. Naïve human CD8 T cells from SIRPγhigh donors were subjected to SIRPG KD and subsequently analyzed for CD45RO expression as shown in Fig. 4A. To evaluate effector function, intracellular levels of TNFα, IFNγ, and granzyme B were measured by flow cytometry in (A) CD45RO− and (B) CD45RO+ subsets. CD45RO+ cells produced significantly higher levels of TNFα, IFNγ, and granzyme B compared with their CD45RO− counterparts, indicating that SIRPG KD promotes functional differentiation of CD8 T cells toward an effector phenotype. HeLa (A–D) and MCF7 cells (E–H) were treated with 30 μM of each indicated STING agonist or DMSO vehicle control (for diABZI) for 24 hours, then harvested in TRIzol for RNA purification and RT-qPCR using TaqMan assays for CCL22 or IFNβ.
