4/4 Our data supports the convergent evolution of a biomolecular condensate. We show that the main pyrenoid assembly protein, SUPA1, has minimal sequence conservation to characterized unicellular algal Rubisco-condensing proteins, binding to a different surface region of Rubisco.
3/4 We provide a unique insight into Rubisco condensation. We combine multi-discipline approaches to reveal the helical folding of Rubisco Binding Motifs upon binding, combined with steric hindrance that halves the available binding sites from eight to four.
2/4 Previous understanding in seaweeds is limited to physiological studies, here we map out a comprehensive spatial atlas of the carbon fixation machinery revealing multiple unique features, for example 68 pyrenoid associated proteins localizing to different sub-domains.
1/4 Until now, seaweeds have proven to be recalcitrant to robust large-scale genetic manipulation. We have developed large-scale fluorescent protein tagging, localizing 160 proteins to provide an unprecedented insight into the spatial organization of seaweed CO2 fixation.
