The tools in OpticalFlow3D are applicable across many length scales, including whole organisms! Measurements of dorsal-ventral motion highlight folding during Drosophila embryogenesis. (4/7)
OpticalFlow3D is designed to apply a Lucas-Kanade-based flow algorithm to 3D images, revealing complex biological motion. Here, flow highlights formation of a contractile actin ring in a dividing cell -- dark blues indicate less motion, while yellow highlights regions of high actin activity. (3/7)
Optical flow measures spatial and temporal changes in intensity, rather than the motion of single object. This allows it to quantify myosin II motion even when the fibers are difficult to segment. In this movie, blue indicates outward motion, while red indicates inward motion. (2/7)
