We find that in mutant worms, sexual maturation is significantly delayed – when wild-type worms are already beginning to mature, l-cry mutants are still immature. So, the worm’s trick to #longevity is to stay longer “pre-puberty”... But what could be molecular mechanisms? [4/7]
Worms exhibit life-long growth, and consistently, in the absence of light and/or upon knock-out of the l-cry light receptor, worms grow to a significantly larger body size. So how about their different life stages, like entry into reproductive phase? [3/7]
We found that knock-out of the sun-/moon light discriminating photoreceptor l-cryprochrome (l-cry; see doi.org/10.1038/s414...; doi.org/10.1073/pnas...) leads to the extension of lifespan in bristleworms by 40%. This is like shifting average human #lifespan from 73 years to 100+ years… [2/7]

The brain also shows dramatic changes when worms enter the #reproductive phase. One of these is the establishment of a molecular state reminiscent of mammalian neuronal stem cell #quiescence – another fascinating cross-reference to vertebrate neural stem cell #modulation @sfb-f78.bsky.social [6/7]
Indeed, when shifted to darkness, wild-type worms showed a marked reduction in proliferation of photoreceptor progenitors. But engineered c-opsin mutants were indifferent to such ambient light changes. This finding was further supported by #scRNAseq analyses of wild-type and mutant brains… [5/7]
A major surprise was Nadja’s observation that early stages of the worm eye’s #photoreceptor cells expressed a ciliary #opsin gene known from #vertebrate visual photoreceptors. We hypothesized that this opsin might mediate an effect of light on proper development of worm eye photoreceptors… [4/7]
