9/10 – Finally, we tried to see what limits this stomatal maturation to the leaf tip. ERL1/2 exprssion show regional differences with more cells showing expression in the hydathode region. In addition, we see that the enlarged size of tip-located hydathode GCs is controlled by ER signaling.
7/10 – Looking at reporter lines we find late stomatal genes normally not present in the embryo. Transcriptomic analysis shows that guard cell genes upregulated in these embryos are normally associated with late stages of guard cell development but that many guard cell genes are still missing.
6/10 – These cell pairs have thick cell walls with a central cell wall thickening that resembles an incomplete pore. So they have many features that cannot be induced by FAMA but still they are not fully mature.
5/10 – Triple mutant seedlings show many more maturing stomata all across the leaf surface but the first maturing stomata are at the cotyledon tip. And surprisingly, they are already there during embryogenesis, as soon as torpedo stage.
3/10 – To understand the state of these cells we use RNAseq to we find that FAMA misexpression leads to upregulation of several, but few, guard cell genes and FAMA targets. Those guard cell genes are normally expressed in very early stages.
2/10 – We next looked more closely at embryos misexpressing FAMA and see that cells on the upper surface of the leaf can progress further and even start expressing SCAP1, even though they still do not form pores. So, leaf polarity influences stomatal maturation potential.
