I'm speculating that one outcome of Prp2 remodeling is a shift in the U2 snRNA (esp. around U2 G20) that allows for Cwc15 peptide insertion. Thus, Cwc15 could "mark" a correctly (?) assembled and activated spliceosome catalytic center.
The take home story is that Cwc15 fine tune's the active site, likely explaining its impact on alternative splicing in organisms ranging from potatoes to cows.
We find that Cwc15 seems primarily to be involved in formation of the spliceosome active site responsible for 5' splice site cleavage as well as promoting that reaction itself. This isn't too surprising given the protein's location proximal to the U2/U6 disnRNA.
Well, this was a weather alert I hadn't seen before... So far about a foot of snow and counting.
